Proposed mechanisms center on modulation of the GABA system, but no single molecular target has been confirmed. Rodent studies report changes in GABA-A receptor expression and in the turnover of serotonin, dopamine, and norepinephrine in several brain regions. Increases in brain-derived neurotrophic factor and its receptor have also been described after repeated administration. These findings come largely from animal models, and the degree to which they describe human neurochemistry remains an open question. The mechanism is best characterized as multi-system and not fully resolved.
Pharmacokinetic data are limited. Like most short peptides, Selank is vulnerable to plasma and tissue peptidases, and its measured half-life in circulation is short, on a minutes scale. The Pro-Gly-Pro tail slows this degradation but does not eliminate it. Intranasal administration is the route described in most reports, with absorption through the nasal mucosa and a hypothesized path into the central nervous system that avoids the blood-brain barrier. Direct measurements of human brain exposure are unavailable, so distribution claims rest on inference from animal work.
Clinical evidence comes mainly from small studies conducted in Russia, several of which were open-label or lacked robust blinding. Reported outcomes include lower anxiety scores, changes in attention measures, and effects on asthenic states following illness. Sample sizes are typically in the tens of participants, and independent replication outside the region is scarce. Reviews published in English generally note the limited methodological quality of the underlying trials. Whether the compound produces clinically meaningful effects under rigorous conditions remains unresolved.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro, frequently abbreviated as TKPRPGP. It was designed as a structural analogue of tuftsin, a naturally occurring tetrapeptide released by enzymatic cleavage of the immunoglobulin heavy chain. The two additional proline residues at the C-terminal end extend the parent chain and change how the molecule behaves in solution. The free peptide has a calculated molecular mass of approximately 751.9 g/mol and is generally supplied as a lyophilised white to off-white powder.
Development work on the compound began in the 1980s and 1990s at the Institute of Molecular Genetics in Moscow, within the same research programme that produced the peptide Semax. Early investigators sought a tuftsin derivative with improved resistance to enzymatic breakdown and with activity in the central nervous system after peripheral administration. Most of the primary literature from this period was published in Russian, a factor that still shapes how easily the findings can be checked by outside groups.
Naming for this compound is not fully standardised in English sources. The spelling Selanc appears in some transliterations, and catalogue entries may instead list the peptide sequence itself as the identifier. Reference material sometimes groups it with other short synthetic peptides studied for behavioural effects, which can create confusion when citations are compared. Distinguishing the exact sequence from related tuftsin analogues is therefore a practical first step when reviewing any dataset or specification sheet.

Selank is a synthetic heptapeptide developed in Russia. Its sequence is Thr-Lys-Pro-Arg-Pro-Gly-Pro, a seven-residue chain built around the natural tetrapeptide tuftsin. Researchers at the Institute of Molecular Genetics of the Russian Academy of Sciences first described the compound in the mid-1990s. The design combined the tuftsin core with an added Pro-Gly-Pro tail, a modification intended to extend the molecule's stability in biological fluids. Published work on the peptide has appeared mainly in Russian-language journals.
Reported activity for Selank centers on anxiolytic and nootropic effects. Russian clinical reports describe use in anxiety and in cognitive or attention-related complaints. Most of this evidence comes from studies conducted by the same research groups that developed the peptide. Independent replication in other countries remains limited, and no major Western regulatory agency has approved the compound for any indication. The gap between local reports and external verification is a recurring point in discussions of the peptide.
Peptide bonds are vulnerable to protease attack, and Selank is no exception. Measured half-life in serum is short, on the order of minutes in several reports, which explains why intranasal administration is the common route described in the literature. Absorption across the nasal mucosa partially bypasses first-pass hepatic metabolism. Quantitative data on human bioavailability remain limited and are difficult to compare across studies.
Lyophilised material kept dry at minus 20 degrees Celsius or colder is the most stable form, and suppliers commonly state a shelf life of two years or more under those conditions. Once dissolved, degradation accelerates through hydrolysis and deamidation, particularly at alkaline pH or elevated temperature. Working solutions are usually divided into single-use aliquots to avoid repeated freeze-thaw cycles. The choice of reconstitution solvent affects both stability and the ionic strength of the final preparation.
The product must be thujone-free as per TTB guidelines, The word "absinthe" can neither be the brand name nor stand alone on the label, and The packaging cannot "project images of hallucinogenic, psychotropic, or mind-altering effects". Absinthe imported in violation of these regulations is subject to seizure at the discretion of U.S. Customs and Border Protection.

In that same month, in clashes between the Comando Vermelho and the militias, 10 people were killed. Residents were threatened by the fighting groups and the president of an association for local residents was kidnapped and subsequently disappeared. On 5 August, one of the leaders of the militia, Carlos Alexandre Silva Cavalcante, known as "Gaguinho", was killed. On 20 August 2008, the militias carried out a massacre which resulted in the death of 7 people in the Carobinha favela in a false flag operation aiming to frame the Comando Vermelho for the massacre. There was also an attempt to enforce the political candidacy of Carminha Jerominho, daughter of Jerônimo Guimarães Filho, alias "Jerominho", the leader of a militia faction. On 5 October, "Mineiro da Cidade Alta" was killed by the militias for the murder of several militia members. From 2007 to 2008, three politicians were arrested for providing support to the militias: Josinaldo Francisco da Cruz (known as "Nadinho de Rio das Pedras"), Natalino José Guimarães and his brother Jerônimo Guimarães. On 9 June 2009, Josinaldo Francisco da Cruz was killed. Despite initially being opposed to drug trafficking, many militia groups (such as the "Liga da Justiça" faction) have allied themselves with cartels like the TCP, while also recruiting many former drug traffickers, informants and ex-convicts into their ranks, sometimes through coercion.

== History == Harith Rajagopalan, MD, PhD, and Jay Caplan established Fractyl Health, initially named Fractyl Laboratories, in 2010. The company's Revita device gained CE marking in April 2016 and became commercially available in the UK in January 2020. In April 2021, the US Food and Drug Administration (FDA) gave Revita breakthrough device designation to treat type 2 diabetes. The company changed its name to Fractyl Health in June 2021. In April 2022, the FDA approved an investigational device exemption trial for Revita. It became available commercially in Germany in early 2023. The FDA issued additional breakthrough device designation to Revita in July 2024 for weight maintenance in certain people with type 2 diabetes or obesity. Fractyl had an initial public offering on the Nasdaq in February 2024. As of February 2025, the company's REMAIN-1 study is evaluating Revita for patients with obesity to support weight maintenance after use of a GLP-1 drug. In January 2025, Fractyl announced it would focus research for Revita on obesity and people on GLP-1s, citing rising demand driven by the popularity of GLP-1s. The company also laid off some employees focused on type 2 diabetes treatment.
). Another factor that impacts total CPU time needed by a simulation is the size of the integration timestep. This is the time length between evaluations of the potential. The timestep must be chosen small enough to avoid discretization errors (i.e., smaller than the period related to fastest vibrational frequency in the system). Typical timesteps for classical MD are on the order of 1 femtosecond (10−15 s). This value may be extended by using algorithms such as the SHAKE constraint algorithm, which fix the vibrations of the fastest atoms (e.g., hydrogens) into place. Multiple time scale methods have also been developed, which allow extended times between updates of slower long-range forces. For simulating molecules in a solvent, a choice should be made between an explicit and implicit solvent. Explicit solvent particles (such as the TIP3P, SPC/E and SPC-f water models) must be calculated expensively by the force field, while implicit solvents use a mean-field approach. Using an explicit solvent is computationally expensive, requiring inclusion of roughly ten times more particles in the simulation. But the granularity and viscosity of explicit solvent is essential to reproduce certain properties of the solute molecules. This is especially important to reproduce chemical kinetics. In all kinds of molecular dynamics simulations, the simulation box size must be large enough to avoid boundary condition artifacts.
=== Theory of cellular division: S. Rosetta === Work by Fairclough, Dayel and King suggests that S. Rosetta can exist in either single-cellular form or in colonies of 4-50 cells, which arrange themselves in tight knit packs of spheres. This was established by performing an experiment involving the introduction of prey bacterium Algoriphagus species to a sample of uni-celled S. Rosetta organism and monitored the activity for 12 hours. Results of this study demonstrated that cell colonies were formed through cell-division of the initial solitary S. Rosetta cell rather than by cell aggregation. Further studies to support the theory of cell-proliferation were done by introducing then removing the drug aphidicolin which serves to block cell-division. When the drug was introduced, cell division stopped and colony formation resulted through cell-cell aggregation. When the drug was removed, cell-division dominated once again.

Duke Nukem Forever had been a project with a troubled development history at 3D Realms, who had created the Duke Nukem series, since sometime prior to 2000. Due to financial difficulties in 2009, 3D Realms was forced to downsize and ultimately lay off most of the development staff. Take-Two Interactive sued 3D Realms for failing to deliver Duke Nukem Forever. Pitchford, who had prior industry relations with many 3D Realms staff including George Broussard, learned that many of the 3D Realms team were still eager to develop Duke Nukem Forever, working out of their homes on what they could. Pitchford negotiated with Take-Two to bring many of the former 3D Realms staff into a new studio called Triptych Games, housed at Gearbox's headquarters, to continue working on Duke Nukem Forever following 3D Realms' closure in 2009. As a result, 3D Realms sold the rights to Duke Nukem and the existing work on Duke Nukem Forever to Gearbox around February 2010. Take-Two and Gearbox subsequently announced in September 2010 that Gearbox would finish production of Duke Nukem Forever. Duke Nukem Forever was released in June 2011, and received negative critical reception on release, with most of the criticism directed towards the unfinished, rushed state of the game. Despite the criticism, the game topped the charts on release and made a profit. 3D Realms had initially sued Gearbox in June 2013 for unpaid royalties over Duke Nukem Forever, but dropped the suit by September 2013, with 3D Realms' founder Scott Miller stating that it was a misunderstanding on its part.
== History == Cyclic guanosine monophosphate (cGMP) research began after cGMP and cyclic adenosine monophosphate (cAMP) were identified as cellular components and potentially involved with cellular regulation. Upon the synthesis of cGMP in 1960, progress rapidly spread in the understanding of regulation and effects of cGMP. Earl W. Sutherland received the 1971 Nobel Prize in Medicine for his work with cAMP and secondary messengers. This award sparked extensive research into cAMP, while cGMP received less attention, with its biological functions largely unknown until the 1980s. During this period, two pivotal discoveries highlighted cGMP's role in cellular signaling: atrial natriuretic peptide (ANP) was found to stimulate cGMP synthesis through the particulate guanylyl cyclase (pGC) receptor, and nitric oxide (NO), identified as the endothelium-derived relaxing factor, was shown to activate soluble guanylyl cyclase (sGC), producing cGMP to mediate vasodilation in smooth muscle cells. Further components involved with the cGMP were also identified such as cGMP-hydrolyzing phosphodiesterases (PDEs) and cGMP-binding proteins. The awarding of the 1998 Nobel Prize to Robert F. Furchgott, Louis J. Ignarro, and Ferid Murad for their discoveries in the NO-cGMP pathway renewed interest in cGMP research with the 1st International Conference on cGMP being held in 2003.

=== Founding === Wilber Hardee (1918–2008) opened his namesake restaurant in Greenville, North Carolina, on September 3, 1960. After a year of success, Wilber decided to look into expanding his restaurant and opening another location so he met with James Gardner and Leonard Rawls to discuss doing so. Shortly thereafter, the first company store was opened in Rocky Mount, North Carolina, in May 1961 by James Carson Gardner and Leonard Rawls at 329 North Church Street in Rocky Mount, known within the chain as building number 1. That location was demolished in 2007 and replaced with a veterans' park named after Jack Laughery, a former Hardee's chairman and military veteran. According to Wilber Hardee, Gardner and Rawls won a controlling share of the company from him during a game of poker. After realizing that he had lost control over his namesake company, Hardee sold his remaining shares to them as well. However, this story has been disputed by Gardner & Rawls, who bought out Wilber Hardee and began establishing franchises in 1961. According to Jack Laughery, CEO of Hardee's from 1975 to 1990, "Leonard put together an organization with relatively little capital. If it weren't for him and Jim Gardner, there wouldn't be anything of Hardee's Food Systems." Rawls and Gardner sold their first franchises to a small group of longtime friends and acquaintances who formed their own companies and over time, built hundreds more franchised locations. Hardee's Food Systems went public in 1963 with Rawls as president.

Reports describe effects on GABA-A receptor expression, monoamine turnover, and neurotrophic factor levels. These are proposed mechanisms drawn mainly from animal models. No single molecular target has been established.
Intranasal delivery is the route described in most published work. It is used because the peptide is degraded quickly once it reaches circulation. Direct evidence of brain penetration in humans is lacking.
Most trials are small, regionally concentrated, and often lack rigorous blinding or placebo control. Independent replication is limited. Reviews in English generally rate the evidence as preliminary.
The sequence is Thr-Lys-Pro-Arg-Pro-Gly-Pro, commonly written as TKPRPGP. It shares the first four residues with tuftsin and carries three prolines in the chain. The proline-rich tail is the main structural feature that separates it from the parent tetrapeptide.
